Why was the file updated? ii. When was the file updated? METHODOLOGICAL INFORMATION 1. Description of methods used for collection/generation of data: Top-down video of a sample undergoing strain on a custom mechanical testing device was processed using custom MATLAB code. The video was broken down into its component frames and the pixels of each frame were analyzed. Pixels with colors belonging to the point of interest were identified and displayed in ESM_1_A.mp4. Pixels belonging to a certain point have their positions averaged to find the geometric center point, and this center point was tracked across all frames for all points of interest. The results of this tracking and the difference between posts attached to samples and unattached posts was quantified to determine post deflection, as demonstrated in ESM_1_B.mp4. 2. Methods for processing the data: Data collected from ESM_1_B.mp4 takes the form of post deflection on either side of the sample in mm, and the displacement of the sample under strain. Post deflection underwent coversion using an experimental calibration in order to obtain force. This measurement of force from the sample was converted to stress using sample geometry, and the displacement of the sample was also converted to strain. This generates a stress-strain curve and a measurement of sample stiffness via elastic modulus. 3. Instrument- or software-specific information needed to interpret the data: MATLAB R2017b was used to execute the custom code. No external packages are required. 4. Standards and calibration information, if appropriate: Plastic 3D printed posts were used as force transducers. An experimental calibration was used to convert deflection in mm to force in N. One calibration methodology was to use PDMS test strips of known material property to correlate deflection and force. The other calibration methodology was to attach pairs of these posts directly to a commercial mechanical testing device, with a strip of paper connecting the post ends where the sample would be. Forces measured by the load cell upon pulling the two posts apart were correlated to the measured translation, which was equivalent to post deflection. As the first calibration method was more time-consuming but more accurate, the second calibration approach was used after scaling the calibration value thus obtained using the first approach. 5.
